Matrixyl (Palmitoyl pentapeptide-4, Palmitoyl-KTTKS) is a widely used biomimetic signaling peptide in anti-wrinkle cosmetics, designed to mimic natural signaling molecules in the extracellular matrix of the skin. It acts as a "matrikine"—a fragment of matrix proteins released during tissue turnover—that signals fibroblasts to initiate repair and rebuilding processes. In vitro studies have confirmed that its core sequence KTTKS potently stimulates the synthesis of type I collagen and fibronectin. In animal wound healing models, Matrixyl formulated in both patches and creams significantly promoted re-epithelialization and collagen deposition (collagen content increased by approximately 65–89%), with delivery mode influencing efficacy. By topically supplementing this signaling peptide, it may partially counteract age-related collagen loss, thereby improving skin elasticity and reducing fine lines.
