MT-1 (Melanotan I), also known as Afamelanotide, is a synthetic 13-amino-acid peptide and a stabilized analogue of α-melanocyte-stimulating hormone (α-MSH). Its core mechanism involves agonistic binding to the melanocortin-1 receptor (MC1R) with four-fold greater potency than endogenous α-MSH, stimulating melanocytes to synthesize eumelanin. Unlike natural α-MSH, which requires UV-induced skin damage for release, MT-1 induces melanogenesis independent of UV exposure while also promoting antioxidant activity and DNA repair. Clinically, it is approved for treating erythropoietic protoporphyria (EPP) and other photosensitivity disorders, with subcutaneous administration improving patients' tolerance to natural sunlight.
