PE-22-28 is a synthetic peptide derived from the cyclic domain of brain-derived neurotrophic factor (BDNF), demonstrating significant antidepressant-like activity in preclinical studies. Its core mechanism involves activating the small G protein Rac1 to promote dendritic spine formation and synaptic plasticity in hippocampal neurons, while also modulating serotonergic system function. In animal models, PE-22-28 exhibits antidepressant efficacy comparable to classical agents but with faster onset, and lacks sedative, cognitive impairment, or withdrawal effects. Due to its low molecular weight, the peptide can cross the blood-brain barrier and be administered intranasally with favorable bioavailability.
