PNC-27 is a synthetic anticancer peptide composed of the HDM2-binding domain of p53 (residues 12–26) linked to a cell-penetrating peptide sequence. Its core mechanism is highly targeted: cancer cells uniquely express HDM-2 protein on their plasma membrane; PNC-27 binds to this protein and induces transmembrane pore formation, leading to rapid extrusion of cellular contents and tumor cell necrosis independent of p53 activity (a process termed "poptosis"). The peptide selectively kills a broad range of solid and hematopoietic tumor cells (including cervical, pancreatic, and leukemia cells) without affecting normal cells. In vivo studies demonstrate effective tumor eradication in pancreatic cancer and melanoma models with no observed off-target toxicity. PNC-27 remains in the preclinical research stage.
