SNAP-8 (Acetyl Octapeptide-3) is a synthetic anti-wrinkle peptide that mimics the mechanism of botulinum toxin and serves as an upgraded version of Acetyl Hexapeptide-8. Its core mechanism involves competitive inhibition of SNARE complex assembly: by mimicking the N-terminal end of SNAP-25 protein, it interferes with vesicular neurotransmitter (acetylcholine) release, thereby attenuating muscle contraction and reducing expression lines. Clinical studies showed that topical Acetyl Hexapeptide-8 could extend the duration of botulinum toxin therapy for blepharospasm from 3.0 to 3.7 months, with a favorable safety profile. However, the peptide has limited transdermal absorption due to its relatively large molecular weight (889 Da) and hydrophilic nature, with only minimal amounts reaching the stratum corneum (~0.54%). It is considered safe for use in cosmetics at concentrations up to 0.005%.
