SNP-9 (not Snap9) is a glycine-serine-alanine repeat peptide derived from silk fibroin hydrolysate, demonstrating significant neuroprotective activity in Alzheimer's disease (AD) research. Its core mechanism is dual-action: it directly reduces toxic oligomeric Aβ levels by co-assembling with Aβ oligomers to form hetero-oligomers, and it binds to calcyclin-binding protein to regulate abnormal tau hyperphosphorylation. Additionally, in blood-brain barrier injury models, SNP-9 modulates the RAGE/NF-κB pathway, reduces TNF-α levels, and ameliorates abnormalities in tight junction proteins (ZO-1, occludin, claudin-5), thereby alleviating Aβ-induced endothelial cell damage.
