Survodutide is a glucagon/GLP-1 dual receptor agonist peptide that synergistically achieves weight loss, glycemic control, and hepatic protection via GLP-1R-mediated appetite suppression and insulin secretion, coupled with GCGR-mediated lipolysis and hepatic fat metabolism. Phase III trials demonstrate up to 16.6% weight reduction at 76 weeks, predominantly from adipose tissue; in MASLD, 84.2% of patients achieved ≥30% relative liver fat reduction and ~60% attained normalization (<5%), with concomitant ALT improvement. Adverse events are mild-to-moderate gastrointestinal, mainly during dose escalation. It holds promise for obesity and MASLD, with long-term cardiovascular outcomes pending CVOT confirmation.
