Retatrutide is an innovative GIP/GLP-1/glucagon triple receptor agonist peptide that achieves potent weight reduction and glycemic control through synergistic activation of three metabolic pathways. Its mechanism involves GLP-1 and GIP receptor-mediated appetite suppression and insulin secretion, combined with glucagon receptor-mediated energy expenditure and lipolysis, producing superior efficacy beyond mono- or dual-agonists. In the Phase III TRANSCEND-T2D-1 trial, patients with type 2 diabetes achieved a 2% reduction in HbA1c and 16.8% mean body weight loss (approximately 36.6 lbs) over 40 weeks, with an adverse event profile consistent with GLP-1 receptor agonists (primarily gastrointestinal).
